Abstract

Recent perspectives on TMD have proposed that they are multi factional problems with structure, function, external trauma, arthritic deterioration and psychological factors as interrelated causes, Despite extensive studies on the relationship between stress and TMD have been conducted, the pathologic mechanism of TMD remains yet to be elucidated. In the present study, to examine whether caspase-3, which is believed to be a stress enzyme, is involved in the pathogenesis of TMD, expression of caspase-3 in the synovial membrane of TMJ was observed by immunohistochemistry
Twenty one Sprague-Dawley rats (8-week-old) were divided into 2 groups restraint stress (18 rats) and control groups 3 rats The rats of restraint stress group were placed In the plastic cages (150 x 50 mm) during the experiment. The rats belonging to stress group were then sacrificed by neck-dislocation on day 0, 1, 3, 5, 7 and 10 of the experiment, the synovial membranes of TMJ were excised and the expression of caspase- 3 10 the membrane was immunohistochemically observed, The results were as follows 1 On day 1, a weak immunoreactivity to caspase-3 was observed in the synovial membrane of the TMJ
2 On day 5, a strong immunoreactivity to caspase-3 was observed 10 the synovial membrane and immunoreactivity was weaker than that observed m the membrane on day 5 and then gradually decreased until day 10
3 The number of synovial membrane cells was decreased with increasing time after day 7 On day 10, a marked decrease 10 number of synovial membrane cells was observed Overall results suggest that stress may cause alterations in the synovial membrane cells, and 1t 1S also suggested that the synovial fluid may be affected by the stress, which subsequently may alter its functions, leaning to path physiological changes of TMJ functions. However, further stuches are necessary to study how the stress acts as a triggering factor ill activate caspase-3 through yet unknown mechanism and influences the neural and hormonal systems.
